The native molecular forms of acetylcholinesterase (AChE) present in adult Drosophila heads were characterized by sedimentation analysis in sucrose gradients and by nondenaturing electrophoresis. The hydrophobic properties of AChE forms were studied by comparing their migration in the presence of Triton X100, 10-oleyl ether, or sodium deoxycholate, or in the absence of detergent. We examined the polymeric structure of AChE forms by disulfide bridge reduction. We found that the major native molecular form is an amphiphilic dimer which is converted into hydrophilic dimer and monomer on autolysis of the extracts, or into a catalytically active amphiphilic monomer by partial reduction. The latter component exists only as trace amounts in the native enzyme. Two additional minor native forms were identified as hydrophilic dimer and monomer. Although a significant proportion of AChE was only solubilized in high salt, following extractions in low salt, this high salt-soluble fraction contained the same molecular forms as the low salt-soluble fractions: thus, we did not detect any molecular form resembling the asymmetric forms of vertebrate cholinesterases.  相似文献

Sera from six ocular microsporidiosis patients and eight individuals with no history of microsporidiosis were assayed by enzyme-linked immunosorbent assay (ELISA) and by Western blot immunodetection. Microsporidia used as antigen include Nosema corneum, Encephalitozoon hellem, Encephalitozoon cuniculi, and Nosema algerae. Three AIDS patients with known E. hellem infections displayed ELISA antibody titers to E. hellem ranging from 1:400 to 1:12,800. Two patients with unclassified microsporidial infections displayed highest antibody titers to N. algerae (1:1,600 and 1:3,200), a mosquito microsporidian which, reportedly, cannot infect man. A sixth patient with a known N. corneum infection displayed the same ELISA antibody titer (1:1,600) to all four microsporidia. Western blot patterns also were variable among the patient sera; however, the most intense and complex antibody-binding patterns corresponded with the higher ELISA antibody titers. Sera from eight HIV-seronegative individuals with no history of microsporidiosis reacted variably to the four microsporidia. These results suggest that diagnosis of microsporidiosis may depend upon direct detection of the organisms using species-specific antibodies or molecular probes rather than conventional serology.  相似文献

The application of correspondence factorial analysis to four types of sediment in the upper reaches of the Loire estuary enabled the significance of environmental factors in the spatial distribution, abundance and seasonal dynamics of the biotic communities to be determined. In coarse sand, the communities are stable but species are few in number and population densities are low. In muddy sediments (from muddy sand to fluid mud) population densities are high but undergo seasonal fluctuations due to changes in the hydrological regime (especially the dissolved oxygen regime). This is true for the composition of the biotic communities. The oligohaline phase, in which the fluvial element is predominant, is characterized by abundance maxima due to freshwater species. The mesohaline phase, in which the marine element is predominant, exerts a restrictive action on population densities; certain species are supplanted by others, their development being governed by the quality of the environment.  相似文献

Summary The mobile genetic element Tn4430, originating from the gram-positive bacterium, Bacillus thuringiensis, and previously described as the Th-sequence, is the first transposon isolated from the genus Bacillus. In the present work a gene (APH-III) conferring resistance to kanamycin was inserted into this 4.2 kb transposon. Transposition experiments showed that Tn4430APH-III could transpose in the gram-negative host Escherichia coli when its insertion functions were supplied by an intact copy of Tn4430. By transposing Tn4430APH-III directly onto pBR322, it was possible to determine the nucleotide sequence of the terminal inverted repeats of Tn4430 and of the target DNA site. Identical 38 bp in inverted orientation are situated at each end of the transposon and there is a direct duplication of 5 bp at the insertion site. Thus, it is clear that Tn4430 is closely related to the transposons belonging to the Tn3 family (class II elements).  相似文献

The study of the migration dynamics of glass eels(Anguilla anguilla) in a small estuary of the French Atlantic coast shows a two-stage sequence: (1) From November to March, the glass eels migrate upstream by using the tidal currents. The estuarine hydrology leads to a natural trapping of migrants in a typical area where the current speed slows down. The location of this zone depends on hydraulic conditions. The greater the tide is the farther upstream this area will be. This phenomenon leads to an increasing catchability of elvers. (2) From April onwards, when the water temperature reaches 10–12 °C, the glass eels swim actively upstream in the estuary. Then, fish concentrate just below the first dam. This behaviour shift shows the beginning of the colonization process of the whole riverine system.  相似文献

The effects of denervation and direct electrical stimulation upon the activity and the molecular form distribution of butyrylcholinesterase (BuChE) were studied in fast-twitch posterior latissimus dorsi (PLD) and in slow-tonic anterior latissimus dorsi (ALD) muscles of newly hatched chicken. In PLD muscle, denervation performed at day 2 substantially reduced the rate of rapid decrease of BuChE specific activity which takes place during normal development, whereas in the case of ALD muscle little change was observed. Moreover, the asymmetric forms which were dramatically reduced in denervated PLD muscle were virtually absent in denervated ALD muscle at day 14. Denervated PLD and ALD muscles were stimulated from day 4 to day 14 of age. Two patterns of stimulation were applied, either 5-Hz frequency (slow rhythm) or 40-Hz frequency (fast rhythm). Both patterns of stimulation provided the same number of impulses per day (about 61,000). In PLD muscle, electrical stimulation almost totally prevented the postdenervation loss in asymmetric forms and led to a decrease in BuChE specific activity. In ALD muscle, electrical stimulation partially prevented the asymmetric form loss which occurs after denervation. This study emphasizes the role of evoked muscle activity in the regulation of BuChE asymmetric forms in the fast PLD muscle and the differential response of denervated slow and fast muscles to electrical stimulation.  相似文献

We have studied the cytoskeletal nature of a brain subcellular fraction previously shown to contain polyribosomes. We have identified the major proteins of this fraction by electrophoretic comparison to a standard cytoskeletal fraction and by immunodetection. These methods have shown the presence of actin, glial fibrillary acidic protein, and neurofilament triplet proteins. We have also studied the effect of various ions and nonionic detergents on the stability of this structure. We have used a cell-free system derived from hamster brain to investigate protein synthesis during experimental phenylketonuria. In such a system the elongation inhibitor emetine impeded translation in extracts derived from both treated and control animals. On the other hand the initiation inhibitor aurintricarboxylic acid showed no effects on protein synthesis activity of treated hamsters, although it was severely inhibiting in controls. Protoporphyria is generally an autosomal dominant disease that is characterized clinically by photosensitivity and hepatobiliary disease and that is characterized biochemically by elevated protoporphyrin levels. The enzymatic activity of ferrochelatase, which catalyzes the last step in the heme biosynthetic pathway, is deficient in all tissues of patients with protoporphyria. In this study, sequencing of ferrochelatase cDNAs from a patient with protoporphyria revealed a single point mutation in the cDNAs resulting in the conversion of a Phe(TTC) to a Ser(TCC) in the carboxy-terminal end of the protein, F417S. Further, the human ferrochelatase gene was mapped to chromosome 18q21.3 by chromosomal in situ suppression hybridization. Finally, expression of recombinant ferrochelatase in Escherichia coli demonstrated a marked deficiency in activity of the mutant ferrochelatase protein and of mouse-human mutant ferrochelatase chimeric proteins. Therefore, a point mutation in the coding region of the ferrochelatase gene is the genetic defect in some patients with protoporphyria.  相似文献

The expression of end-chain sugar residues and of oligosaccharidic sequences has been investigated in chick germ cells at critical stages during the migration, proliferation and sexual differentiation of these cells. Fluorescent lectins and indirect immunofluorescence studies using the NC-1/HNK-1 monoclonal antibody indicate a remarkable control of glycosylation during germ cell embryonal life. Besides a retained expression of glucose/mannose residues, it was found that alpha- and beta-galactose residues, N-acetyllactosamine and N-N' diacetylchitobiose sequences as well as the sulfated trisaccharidic NC-1 epitope were detectable in a stage-specific pattern. Present at a very high density in the cytoplasm and on the surface of the early germ cells at premigrative and migratory stages, the staining for these carbohydrate sequences gradually disappeared when the germ cells settled and proliferated in the developing gonadal primordia. The disaccharide Gal beta 1----3 Gal NAc was exclusively detected in migrating PGCs. In sexualized gonads, acetyllactosamine and/or diacetylchitobiose were similarly reexpressed in both oogonia and spermatogonia. Spermatogonia displayed beta-galactose residues and a high immunoreactivity with the NC1 Mab, indicating modulations in PGC glycosylations related to the acquisition of sexual phenotypes. In addition NC-1 was found to be expressed in the somatic component of the undifferentiated gonad and in the testis interstitial gland.  相似文献
